A simple and efficient method for the preparation of live leukocytes from peripheral blood using the LeukoCatchTM system

چکیده
Leukocytes from peripheral blood (PB) are of great value for diagnosis as well as basic and clinical research. However, no easy, centrifugation-free method is available for the isolation of live leukocytes from blood. We here develop a simple and quick method for the purification of viable leukocytes from whole blood using novel tools, named tLeukoCatch (tip-type) or sLeukoCatch (syringe-type), which is equipped with three Pall filter layers and captures leukocytes but not red blood cells (RBCs) in whole blood. Indeed, we showed that several million leukocytes per mL (~35% of the recovery rate) were captured and eluted from whole blood. The number of contaminant RBCs decreased from several million to several thousand. When mouse blood was hemolysed, almost all of the lysed RBC fragments were removed by passage through sLeukoCatch. Optical microscopic observation confirmed that the recovered leukocytes were sufficiently healthy to respond to growth stimuli. Efficient leukocyte recovery was also confirmed for hemolysed human blood. These results suggest that the LeukoCatch system is useful for bedside diagnosis and basic research with blood samples.

Effect of famotidine on radiation induced apoptosis in human peripheral blood leukocytes
ABSTRACTBackground: Radioprotective effects of famotidine, an antagonist of H2 receptor clinically used for peptic ulcer treatment, was previously shown on radiation-induced micronuclei and chromosomal aberration in human peripheral blood lymphocytes and mouse bone marrow cells. This study was conducted to investigate radioprotective property of famotidine against radiation induced apoptosis i...
متن کاملSEQUESTRATION OF LEUKOCYTES BY THE ISOLATED PERFUSED RAT LUNG
The sequestration of rat leukocytes and bone marrow cells by the lung vasculature was studied using an isolated perfused rat lung preparation. The passage of latex particles of 7.6 µm in diameter and non-hematopoietic cells through the lung blood vessels was also studied. Leukocytes and bone marrow cells were reversibly sequestered from circulation, whereas, latex particles and non-hematop...
متن کاملConsumption of antioxidant dietary agents, curcumin and vitamin C, protects cellular DNA from gamma-radiation
Background: Exposure to ionizing radiation results in genotoxicity and the unrepaired lesions in cellular DNA results in cell cycle arrest, reproductive death, interphase death, division delay, chromosome aberrations, mutations, etc. leading to the intensive destruction of cells and violation of their proliferative capacity there by adversely affecting the mammalian system. Since ionizing radia...
متن کاملRadioadaptive response in peripheral blood leukocytes of occupationally exposed medical staff with investigation of DNA damage by the use of neutral comet assay
Background: ˝Radioadaptive Response˝ is well-documented phenomenon appeared in low dose ionizing radiation received in vitro and in vivo. Occupational exposure has always been a great concern for radiation workers therefore this study was performed to study radioadaptive response in terms of residual DNA double strand breaks as an endpoint in peripheral blood leukocytes of occupationally expose...
متن کاملAssessment of mitochondrial DNA copy number in peripheral blood leukocyte of opiate abusers and healthy individuals
Background: Based on the studies, variation in the mitochondrial DNA (mtDNA) copy number in peripheral blood leukocytes is associated with increased susceptibility to diseases including cancer. Opiate abusers are at high risk for diseases. In this study, we measured the mtDNA copy number in peripheral blood leukocytes in a group of opiate abusers compared with those in healthy individuals. Met...
